The human 26S immunoproteasome is a native, ATP-dependent multiprotein protease composed of a 20S immunoproteasome core associated with one or two 19S regulatory particles. Its catalytic core is enriched in the immunoproteasome subunits β1i/LMP2, β2i/MECL-1 and β5i/LMP7, which distinguish it from the constitutive 26S proteasome and alter its proteolytic cleavage preferences. The complex participates in regulated intracellular protein degradation and the generation of peptides for MHC class I antigen presentation.
Our 26S immunoproteasome is isolated directly from human spleen tissue by fractionation, without recombinant expression or affinity tags, preserving the complex in its native state.
